Can I determine this microstructure grain size?

Dear all,
I have a steel bend pipe, material: ISO 3183-3.
The location for microstructure and grain size determination (according to E112) was taken at the mid bend inner of the pipe. From the picture attached, it seemed to be tempered martensite with disperse ferrite.

If this is the case, do I need to perform a heat treatment (according to E112) before I can determine the grain size? Thanks!

No, not for grain size. If you perform a heat treatment on this bend it will alter the current microstructure and mechanical properties.
 
You can determine the grain size but you'll need to increase magnification and you need to be more familiar with metallurgy than appears by your post.

Agree with metengr...heat treatment will change the microstructure (grain size and intergranular observations)
