fabrication of spindle with bolted end

I'm fabricating the spindle with bolted end to tighten the parts.

I have doubt about

has thread size and no. of thread any impact on operations?

for 1.25inch dia rod would to threaded its end.

Kindly suggest me how many number of thread on shaft and size of bolt.
