I will research a initial topology of laminar composite. How dependent on the initial structure is the optimized solution? how the simulation could look like. What different initial states are interesting?

Topology optimization algorithm only removes unnecessary material from the model so you need some reasonable initial guess. Various constraints will be important as well. For example you can restrict the optimization so that no material is removed in particular regions, symmetry is conserved or selected manufacturing constraint are included. If you don't want the software to rely on your initial geometry then consider using generative design. This interesting technique is available in selected CAx programs.
