Assume you are analyzing a vessel on 4 legs. The moment at the base of the
legs is 10000 ft-lbs due to seismic condition. The large nozzle in the top head has a
nozzle load of 10000 ft-lbs. Would you design the vessel support legs for a total
of 20000 ft-lbs?

Table 5.3 od ASME VIII Div 2 will point you in the right direction.
 
Yes, in-service loads should typically be applied in conjunction with environmental ultimate design loads (think wind, seismic, snow etc.) although these enviro loads are generally not applied concurrently.
