Height of water seal

I would like to know the height of water seal required in a floor trap which is subjected to a vacuum pressure of
-50 pa(the trap is located in a lab which is under negative room pressure)

50 Pa corresponds to 5 mm of water column. That means the given negative pressure can hold a water column of 5 mm height and rest of the water will be drained. Generally the U Seal for floor drains will be of 1.5" to 2". So no problem in your case.
