Does closed vents = closed returns?

Hi y'all

I've recently moved into an old school house (built around the 1870s), and an oil furnace was added at some point along the way. The house is 2 storeys, with the upper floor being around 24' x 31' x 12 1/2'. This is quite a large room, and as such, is difficult to heat. The previous owners have a shut off lever in the basement to close the heating vents that run upstairs. I'm wondering, if I am not heating the upstairs, should I block the cold air return too?

The difference in the floors is about 20C compared to 10 - 14C (depending on the cold temperatures outside)

Any comments are greatly appreciated.
