Is there any effect if we fill up oil through the air vent?

What air vent?

You need to annotate that diagram as it looks like a closed chamber to me.

Remember - More details = better answers

Do you have a problem with following manufacturer's warnings and directions? Bad idea! What vent? Any vent.

Oil coating the vent pipe, or remaining in the vent may combine with dirt and microbes, turn to sludge and/or simply dry, blocking and sealing in any vapors from escaping.



 
Using this oiler, you have no visual indication of the oil level in the housing. If you fill the housing from the vent, you may overfill it and not know. Simply remove the oil bulb and watch the oil level in the cup while filling and you avoid this problem. It is a good idea to fill the housing through the oiler cup just to verify it is piped correctly and there is no blockage in the line between the oiler and the housing.

You cannot fill the glass oil bulb (reservoir) by adding oil through the housing vent. Oil can't push up into the bulb unless it is turned over. Maybe that is what the note means.

That too. There is no guarantee that oil will fill the reservoir, as no vapor can be forced out through the vent while it is being filled.

 
It is also possible they are referring to the equalization tubing on the pressure equalized, non-vented design. That is usually very small tubing (0.25 inch). If you added oil through that tubing, it could form a liquid trap that would prevent the oiler from properly equalizing the pressure in the oiler to the pressure in the housing. Why would anyone try to add oil through 1/4 inch tubing?

The original question answers that question. Why would anyone try filling through any kind of vent? If they're thinking about that, any olho will do.
