what is the most important things to consider in selecting the right bolt and nut.

not really, i'm working in industrial sector, but have no experience in designing machine or its parts
 
OK, I suggest you start by by reading some of the many threads here on bolting. There are MANY factors involved with bolted designs, and you really shouldn't get any of them wrong for critical applications. Here's a few:

1. Corrosion concerns
2. Clamping force required
3. Size restrictions
4. Type of loading (fatigue cracking concerns)
 
As simple as this sounds it still gets missed so it's worth repeating.

On Metalguy's item 3, make sure you've got enough room for the tooling/person that will be used to tighten it.
