I need to design one Macro who create layers, in ascending order, and create the name according to an alphanumeric pattern controlled and incremental (f.e. spline_01, spline_02, etc). In each one of these layers we need insert a spline (diferent XYZ points in a txt file).
If when you go to create a layer you detect that in this layer contains some objects, or if it is already named, the Macro should jump to the next one.
The ideal, it will be to create the Macro like a loop and parameterized.

A GRIP program will do that job and be easier to control and modify.

I can't help but ask why you need to do this in the first place? After all, with only 256 layers, you could soon be out of places to put Splines. Perhaps it might be better to just give them unique names and Hide them and then use some Macro/Program to Show the needed Spline.
